Within the laboratory, activities include the purification of total protein extracts, enrichment of peptides containing post-translational modifications, one- and two-dimensional electrophoretic and chromatographic separations, image analysis of 2D maps, western blotting, and protein identification and quantification by mass spectrometry coupled with bioinformatic analysis.

Equipment

  • Proteomics platform based on the 2DE (two-dimensional electrophoresis) approach: IEF separation (IPGphor, GE Healthcare, and PROTEAN® i12™ IEF system, Bio-Rad), SDS-PAGE electrophoretic separation (PROTEAN II XL cells for six large gels, Mini-PROTEAN cell, Bio-Rad), image analysis (ImageMaster, PDQuest)
  • Western blot system (for mini and large gels, Bio-Rad) with Lycor Odyssey imaging system
  • LXQ linear ion trap (Thermo Fisher): linear ion trap mass spectrometer with ESI source. The instrument is equipped with an HPLC system (MS Pump Plus, Thermo Fisher) and an autosampler (large-scale equipment)*
  • Cluster consisting of 12 CPUs dedicated to proteomic data analysis
  • Bioworks software for protein identification; in-house bioinformatic analysis tools
